1) My one employee thinks I am a miser.

Is that sentence grammatical?
Is it meaningful?

I think that sentence in unacceptable in formal English,
but might be acceptable in informal English. I am not sure at all.

If it is acceptable, does it mean:
My only employee thinks I am a miser.

Could it mean: One of my employees thinks I am strange.

Gratefully,

Yes.

I think I'd label it as fairly formal.

Yes. His name is Bob Cratchit.

> Could it mean: One of my employees thinks I am strange.

No, not even if 'strange' is taken to mean 'a miser'.
